LIBRARY PREP

Target enrichment and library preparation

Focuses sequencing on the genes that matter, by hybridisation capture or multiplex amplicons.

Immunoassays explained

The four ELISA formats

All ELISAs use an antibody to catch a target and an enzyme to turn a colourless substrate coloured. They differ in what is fixed to the plate and what is measured.

Antigen on the plate; one enzyme-labelled antibody detects it.
Antibody from the sample binds coated antigen; a labelled secondary antibody detects it. Used for serology.
Two antibodies grip the antigen from both sides. The most specific format; used for cytokines and biomarkers.
Sample antigen competes with labelled or coated antigen: less colour means more analyte. Used for small molecules such as mycotoxins and drug residues.

Sequencing in depth

Long reads or short reads?

Short-read sequencing is the workhorse for SNVs, panels and ctDNA. PacBio HiFi long reads resolve the structural variants, repeats, methylation and phasing that short reads miss. Many labs need both.

REFERENCE LOCUSrepeat regionSHORT READS · 150–300 bpreads inside the repeat map ambiguously; the region stays unresolvedPACBIO HIFI READ · 15–20 kbone molecule anchors in unique sequence on both sides: structure, phase and 5mC in a single read

What long reads add

Structural variants

Deletions, inversions and translocations are read end to end.

Repeat expansions

Long reads anchor in unique sequence on both sides of a repeat.

Native 5mC

Methylation is read from the kinetics of the same run.

Phased haplotypes

Variants across a gene are linked on one molecule, which matters for HLA.

Full-length isoforms

Kinnex and Iso-Seq read complete transcripts and fusion partners.

Strain-level microbiome

Full-length 16S separates species that V3–V4 amplicons cannot.
